I have a Q Max coolant reroute. Rather than going through the conventional way of thermostat housing --> Turbo --> Mixing Manifold, would I have any issues by using the Thermostat block off plate in the front of the engine (M16x1.5 to -4AN) --> Turbo --> Rear of engine to the QMAX housing (1/8npt to -4AN)? I'm looking at the M-Tuned coolant flow diagram and if i'm looking at it correctly, I shouldn't have any issues doing this.

No. To run head to head, one in front, one in back will not work correctly. Don't do that.
End at the mixing manifold. Start from either the block-off plate, or the QMax housing.
With either of those recommended routings:
During operation, the flow direction will, again, in either case, go from the head, through the CHRA, and back to the mixing manifold.
After shutdown, the source of flow will be natural convection caused by CHRA heat, and move from the mixing manifold, up to the turbine CHRA, and then to the head.
